The original TomTom GO comes with a 128Mb SD card (not mini-SD). My boss asks if he can upgrade it to 512Mb or 1Gb?

Yes, TT is desined to work with SD's up to 4GB. (which I don't think actually exist yet)

A lot of people are using 1GB cards with their TT GO nowadays.

Thanks. Is it a case of re-uploading the maps? Or can you just copy them from one card to another?

Also, this is an original Tomtom GO, not the 300 or 700 or whatever?

You can choose. But the easiest way is indeed simply copying the entire contents of the SD card.

Makes no difference. All the Go's work alike. Maybe with the exception of the 700 who uses a HD instead of SD cards, and thus already has all the maps of Europe pre-installed.
